5-Amino-1MQ (5-amino-1-methylquinolinium) is a small-molecule nicotinamide N-methyltransferase (NNMT) inhibitor widely used in research on NAD⁺ metabolism, adipocyte biology and metabolic disorders. 5-Amino-1MQ emerged from medicinal-chemistry efforts to create more potent NNMT inhibitors than the early probe 1-methylquinolinium. Quinoline-derived analogs such as 5-Amino-1MQ showed markedly improved NNMT inhibition, good cell permeability and robust effects on adipocyte methyl-nicotinamide levels, prompting their use in obesity, metabolic syndrome and NAD⁺-related research programs.
5-Amino-1MQ is a quinolinium derivative bearing an amino group at the 5-position and a methyl group on the ring nitrogen. The cation typically has the molecular formula C₁₀H₁₁N₂⁺ with a molecular weight of ~159.2 g/mol (counter-ion dependent). Its planar aromatic scaffold and positive charge support binding in the NNMT nicotinamide site.
In differentiated adipocytes, 5-Amino-1MQ lowers intracellular 1-methylnicotinamide in a concentration-dependent manner, increases NAD⁺ levels and suppresses de novo lipogenesis without significantly inhibiting closely related methyltransferases. In rodent models of diet-induced obesity, NNMT inhibition with 5-Amino-1MQ has been associated with increased energy expenditure, reduced fat mass and improvements in metabolic parameters, supporting NNMT as a target in obesity and cardiometabolic research.
Put simply, 5-Amino-1MQ lets researchers test what happens when cells and animals waste less NAD⁺ and shift fuel use away from fat storage. Early work shows promising shifts in metabolism in lab and animal studies, but these findings are exploratory and not evidence of proven benefits or safety in humans.
